  • a Whole-blood samples were incubated for 5 min at 37°C with PBS or with the following TLR agonists: LPS (10 ng/ml; TLR4); MALP-2 (10 ng/ml; TLR2/6); Pam3CSK4 (500 ng/ml; TLR1/2), R-848 (10 μg/ml; TLR7/8); or CpG-DNA (100 μg/ml; TLR9). Phospho-IRAK-1 content was then measured by flow cytometry on methanol-permeabilized cells as described in Materials and Methods. Results are expressed as mean ± SEM (n = 3; each experiment was performed with a different healthy control).
