  • a The IL-10-NOD mice, of N3 and N4 backcross generations, homozygous or heterozygous for the scid mutation, were generated by backcrossing of the IL-10-BALB/c mice with NOD-scid mice for 3 to 4 generations. The resulting mice were typed for scid homozygosity (absence of T and B cells) by FACS analysis and for the IL-10 transgene by PCR. The results are represented as pooled data. Mice were bled at weekly intervals by retrobulbar puncture for determination of BG concentration. Mice were considered diabetic when the BG lvels were >300 mg/dl. Control mice are transgene negative littermates. The IL-10-NOD-scid mice (n = 13) were obtained upon screening a total of 92 pups belonging to ten different litters. The wild-type IL-10-NOD mice (n = 12) were obtained upon a screeing of 47 pups belonging to five different litters.
