OT-II T cell and DC interaction.
OT-II Tg CD4+ T cells were added to overnight LPS/αCD40 matured CD11c+ CD80-eCFP splenic DC pulsed with 5 μM OVA. TCR was labeled with H57 Fab. This is an 11 min movie. CD80 is shown in green and TCR in red.
Supplemental qtvr 1 (MOV, 7.41 Mb)
-
3D view of T cell-DC IS corresponding to Figure 4A. anti-CD80 is shown in green, anti-TCR in red, and anti-LFA-1 in blue. The interface is demarcated by the numerous CD80 clusters interspersed with a few small TCR clusters. Some larger TCR clusters appear to be in invaginations to the side of the IS.
Supplemental qtvr 2 (MOV, 9.17 Mb)
-
3D view of T cell-DC IS corresponding to Figure 4B. CD80-eCFP is shown in green, anti-TCR in red, and anti-LFA-1 in blue. The T cell is interacting with a process from the DC and the DC body through a dense network of LFA-1 clusters interspersed with CD80-eCFP and TCR clusters.
Supplemental qtvr 3 (MOV, 8.79 Mb)
-
3D view of T cell-DC IS corresponding to Figure 6B 90s. CD80-eCFP is shown in green and anti-TCR Fab in red.
Supplemental qtvr 4 (MOV, 9.07 Mb)
-
3D view of T cell-DC IS corresponding to Figure 6B 210s. CD80-eCFP is shown in green and anti-TCR Fab in red. Comparison of qtvr 3 and 4 show how a TCR cluster appears to emerge in area that had previously been dominated by CD80 clusters.
