Supplemental Figure 1. Detection of IFN-γ response to Mamu-A*02 binding peptides by PBMC from Mamu-A*02+ SIV-infected macaques (A) 95061, 97086, AJ11 (B) 96078, 90098, 96115, (C) 98015, and 98020 using the ELISPOT assay. Freshly isolated PBMC from various Mamu-A*02+ SIVmac239-infected macaque were tested with 75 Mamu-A*02 peptides in ELISPOT assays. Shown are the 21 responses that were positive in at least one Mamu-A*02+ macaque. Mean values and standard deviations from triplicate wells were calculated for each assay. Red bars indicate positive responses, while blue bars are negative responses. NT indicates not tested. Background (the mean of wells without peptide) levels were subtracted from each well. Responses <50 SFC per 1 x 106 cells are not considered positive because these counts are not significantly above background. Assay results are shown as SFC per 1 x 106 cells. Responses to Con A (positive control) were always greater than 1000 SFCs per 1 x 106 cells.
Supplemental Figure 2. Sequence analysis of 14 novel Mamu-A*02 CD8+ T-cell epitopes in chronic SIVmac239 infection. The 14 Mamu-A*02 restricted CD8+ T-cells epitopes were directly sequenced from cell-free plasma from SIVmac239-infected Indian rhesus macaques at least 30 weeks post-infection. Codons identical to the wild-type sequence are shown as dots. Complete amino acid replacements are shown as uppercase single-letter amino acid codes; mixed populations are shown as lowercase single-letter amino acid codes. "X" indicates that several amino acids could be encoded. Italicized animals (2161 and 2162) are Mamu-A*02-.
