Fluorescent beads and OVA taken up by an iris cell. A 3-dimensional reconstruction of an iris APC derived from a Z-stack of confocal images. The cell had taken up green fluorescent OVA and red beads. Beads not associated with cells taking up fluorescent green OVA are also in the image.

APCs do not leave the iris 5 hours after antigen challenge. An hour time-lapse recording of BALB/c iris was started 5 hours after the injection of green OVA and red beads into the anterior chamber. No significant movement of green OVA labeled cells is observed. Some beads not associated with green OVA-labeled cells move but are not seen leaving the iris (two are marked with arrows prior to the start of the movie).

An hour time-lapse recording of BALB/c iris starting 4 hours after the injection of green OVA and red beads into the anterior chamber and 7 hours after LPS injection intravitreously. No significant movement of green OVA labeled cells is observed. Beads not associated with green OVA labeled cells move but are not seen leaving the iris.

An hour time-lapse recording of BALB/c iris starting 46 hours after the injection of green OVA and red beads into the anterior chamber and 49 hours after LPS injection intravitreously. No significant movement of green OVA or red bead labeled cells is observed.

No beads moving in the iris of a neutrophil depleted mouse 5 hours after antigen challenge. Five hours after the injection of green OVA and red beads into the anterior chamber, an hour time-lapse recording of the iris of BALB/c mouse depleted of neutrophils by anti Ly6-G antibody. No significant movement of green OVA-labeled cells is observed. A number of free-floating beads are discernible in the aqueous humor, but no beads moving within the iris are observed.
