Supplemental Data for "Transient Association of Ku with Nuclear Substrates Characterized Using Fluorescence Photobleaching" by William Rodgers, Stephen J. Jordan, and J. Donald Capra
Supplement 1

The FRAP experiment of the untreated sample expressing Ku70-GFP shown in Fig. 2A. The first frame is a prebleach image, and the following frames were collected every 1.7 s beginning 1.4 s following a 0.5-s photobleach pulse.

Supplement 2

The FRAP experiment of the fixed sample expressing Ku70-GFP shown in Fig. 2A. The time intervals between each frame are the same as those in Supplement 1.

QuickTime Video 2

Supplement 3

A FRAP experiment using a HeLa cell expressing Ku86-GFP. The time intervals between each frame are the same as those in Supplement 1.

QuickTime Video 3

Supplement 4

A FLIP experiment using a HeLa cell expressing Ku70-GFP. The movie consists of a prebleach image, and each subsequent frame was collected following a 5-s photobleach pulse. The cell adjacent to the bleached cell was not bleached during the experiment, thus showing that photobleaching was due to the laser pulses alone.

Supplement 5

A FLIP experiment of a HeLa cell expressing Ku70-GFP. The sample was pretreated with wortmannin and irradiated prior to photobleaching. Each frame was collected following 5 s of photobleaching.

Supplement 6

A FRAP experiment using a HeLa cell expressing Ku70CD-GFP. The time intervals for each frame were the same as in Supplement 1.
