These movies show the Ca2+ and morphological responses induced by APCs in T cells overexpressing kinase-defective forms of Lck and Itk. The level of [Ca2+]i in T cells is displayed in color ranging from blue (50-100 nM) to red (600-800 nM).
Movie 1: Ag-induced morphological and Ca2+ responses in one wild-type T8.1 cell. Please note the small contact interface which occurs before the Ca2+ increase. Frame interval, 5 s.
Movie 2: High magnification of T8.1/Ag-pulsed APC interaction. Morphological changes are characterized by extended lamellipodia and intense membrane ruffling. Frame interval, 4 s.
Movie 3: Ca2+ and morphological responses of three Lck-KD cells interacting with Ag-pulsed APCs. Two of the three T cells do not respond. However, one T cell displays marked morphological and Ca2+ responses. Frame interval, 11 s.
Movie 4: Ca2+ and morphological responses of two Itk-KD cells interacting with Ag-pulsed APCs. One T cell displays a weak Ca2+ response despite extensive shape changes. Frame interval, 12 s.
