Alteration of the Specificity of Lectin by Chemical Modification1

Shoji Matsubara and William C. Boyd

From the Boston University School of Medicine, Boston, Massachusetts

Abstract

What is thought to be the first instance of the modification of the specificity of a lectin by chemi-treatment is reported. Coupling diazotized p-aminophenyl - N - acetyl - D - galactosaminide to anti-A lima bean lectin greatly increased the agglutinating power of the lectin for group B cells without significantly altering its agglutinative activity for A1, A2, or O cells. The question is raised; do these observations possibly point to a role of carbohydrates in the specificity of lectins (and possibly of antibodies)?
