The Synthesis of -Globulin by Rabbit Lymph Node Cells¹

Abstract

Experiments were performed to test the possibility that the three papain fragments of rabbit -globulin are synthesized separately in lymph node cells. Cell suspensions were incubated with tritiated leucine for periods between 30 sec and 24 hr. Incorporation of the label was stopped at the desired time by dilution with unlabeled L-leucine. The cells were harvested, washed and homogenized, and the labeled intracellular -globulin was isolated in the presence of carrier, cleaved with papain, and examined for the distribution of radioactivity among the three fragments. Essentially uniform labeling was found in all experiments. It is therefore unlikely that the papain fragments form separate pools from which the whole -globulin molecule is assembled.
