Studies on Organ Specificity

XII. Fractionation of Hog Thyroid Extract. Preparation of Thyroglobulin and Thyralbumin1

Sidney Shulman2 and Ernest Witebsky

Department of Bacteriology and Immunology, University of Buffalo School of Medicine, Buffalo, New York

Abstract

1. By means of precipitation with ammonium sulfate, the major globulin and albumin fractions of thyroid extract have been isolated. The required levels of precipitant are, respectively, 1.60 to 1.70 M and 2.50 to 3.00 M. Each fraction seems highly purified by ultracentrifugal analysis.
2. The method is readily reproducible, and does not require any special equipment. It is adaptable to handling of large quantities of extract solution.
3. A new protein fraction of thyroid extract, thyralbumin, has been isolated and partially characterized.
