Concentration and Purification of the Toxin of Streptococcus Hemolyticus with Acetone

Abstract

In recent years many attempts have been made to develop a simple and trustworthy method of concentrating and purifying the toxic substances in sterile filtrates of cultures of streptococci. Various methods have been followed but in general they consist of precipitating the protein-toxin fraction and then purifying by dialysis or ultrafiltration. Huntoon (1) used ammonium sulfate and acetic acid. Henry and Lewis (2) precipitated the toxin with alcohol. Hartley (3) dialyzed through collodion and then precipitated with acetic acid. Pulvertaft (4) precipitated whole broth culture with alcohol. Ando (5) precipitated his toxin with absolute alcohol. Shinn (6) claims to have secured the best results with acid alcohol. It is evident from the results obtained with these different methods that the toxic substances of the streptococcus can be concentrated and, to a certain extent, purified in the process.
