Further Purification of the Glycoprotein that Induces Nephrotoxic Antibody: Isolation of the Active Polysaccharide Fraction Mainly Composed of Glucose

Abstract

Experiments that were done to determine the function of the protein and polysaccharide moieties of a highly purified renal glycoprotein revealed that the antigenic activity that induces nephrotoxic antibody relates more closely to the polysaccharide moiety.

By using the 90% phenol extraction method, the active polysaccharide fraction which possesses antigenic potency was extracted from renal glycoprotein that was shown to contain no nucleic acid. In this highly purified antigen (glycoprotein) the polysaccharide moiety was shown to consist mainly of glucose and the protein content was remarkably low, without any decrease in the antigenic activity that induces nephrotoxic antibody.
