Assignment of Direct and Facilitated Hemolytic Plaques in Mice to Specific Immunoglobulin Classes

Abstract

The kinetics of appearance of hemolytic plaque-forming cells in immunized mouse spleens has been studied with the aid of immunoglobulin specific antisera and myeloma protein inhibitors. "Facilitated" or "indirect" plaques were developed with antisera specific for {gamma}1, {gamma}2 and {gamma}M, and for {kappa} light chains. Facilitation with anti-{gamma}M antiserum suggests the presence of two classes of mouse macroglobulin, one responsible for direct plaques and the other requiring an anti-globulin reagent in order to produce lysis. The previously held concept that all indirect plaques represent "7 S" antibody is no longer tenable.
